Generic Lyssa builds are based on being able to attack as quick as possible in as short time as possible, so you'll need an IAS and some activation time skills. With that said, this is usually a good starting point:
[skill]Mystic Sweep[/skill][skill]Eremite's Attack[/skill][skill]Protector's Strike[/skill][skill]Heart of Fury[/skill][skill]Avatar of Lyssa[/skill]
From there, you can just insert whatever else you feel like. Some good options would be energy management ([skill=text]Zealous Renewal[/skill], although it isn't really needed if you have a Zealous Scythe), self defense ([skill=text]Conviction[/skill] and/or [skill=text]Mystic Regeneration[/skill] are good for starters), or more attack skills as you see fit ([skill=text]Victorious Sweep[/skill] and Zealous Sweep are decent options).
I haven't played a Melandru build in PvE at all though, so I can't help you there.
Also, Aura of Holy Might is essential to all PvE Dervish builds unless you absolutely need every single skill in the build you're running. And yes, it's that good XD.
Typically, runes would either go as:
Scythe Mastery: 12+1+1
Mysticism: 12+1
Or if you're running Earth Prayers as well, this is pretty standard:
Scythe Mastery: 11+1+1
Mysticism: 11+1
Earth Prayers: 8+1
For HM, I would reccomend staying away from Avatar of Lyssa due to the faster activation times that all enemies have, which leads to less hits benefitting from the damage bonus that it gives.

"I am the strongest" (or Rez)
Lyssa
Eternal Aura
Heart of Fury
Aura of Holy Might
12+1+1 scythe
12+1 myst
lotsa norn points
zealous 15^50 of enchanting scythe
the attack skills are interchangeable for things like victorious sweep, protector's strike or bull's strike.

Would it be better to replace wild blow with Radiant Scythe?

Naw, a guaranteed critical hit on a scythe that cannot be blocked and removes a stance is better than a maximum of +53 dmg (if you are running a sup scythe rune and hood). The 2 second faster activation time on radiant scythe doesn't really make up for it either. Of course, wild blow makes you take warrior, but it really is a good secondary for dervs, see the other warrior skills mentioned in this thread.

naw I wouldnt drop wild blow. you could replace on of the other attack skills though. you mostly end up getting to lazy to time them during spells anyways.
as a sidenote, Radiant scythe fits easily in any build with a zealous scythe, you shouldnt need the bonus from lyssa to have your energy around 20.

I don't think Vital Boon is worth having, but your choice I suppose. At least it feeds Mystic Sweep a bit. If it were up to me and I wanted a defensive-ish skill I'd use Great Dwarf Armor instead though, since it helps survivability more and still has 100% uptime.
Also, unless you don't have it, AoHM is better than HoHF.

Basic build I run with PvE or on a Hero. Enough enchantments for +10 regen and max Mystic Sweep. I like a balance of offense and defense, this build survives pretty much anything in any zone and does good consistent damage. With 45 energy you can cycle your two sweeps non stop while maintaining your defenses.

I'd personally say the only required skills that are absolutely needed with [skill]Avatar of Melandru[/skill] are [skill]wearying strike[/skill] and [skill]Eternal Aura[/skill].
[skill]Aura of Holy Might[/skill] should be on every dervish bar. [skill]heart of fury[/skill] should also be there unless you have some other IAS skill.
